Assay Detail The membrane permeability of odoamide derivatives was evaluated using a PAMPA assay (Corning®Gentest™ Pre-coated PAMPA Plate System) by the support service from the Drug Discovery Initiative in the University of Tokyo, according to the manufacturer's protocol with a slight modification. Phosphate buffer solution (0.1M, pH 7.4) was used as a donor and acceptor buffer, and initial donor solutions were prepared by diluting DMSO solutions in phosphate buffer solution to be 10 µM. Permeability of the compounds was calculated using the equation described in the previous study (https://link.springer.com/article/10.1007/s11095-007-9517-8). The peptideconcentration was quantified by LC-MS/MS analysis. No significant peak(s) with the identical mass number were observed during the incubation (37 °C, 4 h).
